Drug intelligence / Profile preview
[64Cu]NOTA-TVS-W-PSMA dimer is a dimeric, prostate-specific membrane antigen (PSMA)-targeted radiopharmaceutical developed by the University of North Carolina at Chapel Hill and licensed to Tripill Biotech. Built upon a tri-vinyl sulfone (TVS) scaffold, the agent incorporates a tunable tryptophan (W) amino acid linker between the PSMA-targeting ligand and the vinyl sulfone moiety to optimize pharmacokinetics. Radiolabeled with copper-64 (64Cu), this dimeric PET tracer is designed for the diagnostic imaging of PSMA-positive prostate cancer. In preclinical studies, the tryptophan linker significantly reduced off-target kidney and salivary gland uptake while maintaining high tumor retention and specificity, addressing the renal and glandular toxicities associated with conventional monomeric PSMA-targeted agents.
